Can I change the file manager used for STRG+UMSCHALT+E? I use the Speedcommander instead Windows Explorer. In the favorites I have added it and can be used.

No. This command uses a dedicated Windows routine to start the standard file Explorer.  Use a Favorite and assign a keyboard shortcut to it for the same effect.
